Mammalian cell lines

Normal mouse and human cell lines (e.g. fibroblasts, keratinocytes, endothelial cells, etc). A big collection of human and mouse cancer cell lines along with cancer cell lines being chemoresistant to chemotherapeutics (doxorubicin).

Fly model

More than 250 transgenic lines covering the entire spectrum of proteostatic, mitostatic, metabolic and DNA damage repair pathways, as well as models of several human diseases.

Mouse model

Two transgenic lines, and models for tumorigenesis-related assays (see also below).

Participation in clinical trials

Bioassays in blood cells (see also below).

High throughput screening of drugs/natural products

Numerous cell-molecular biology assays (e.g. cellular senescence, inflammation, angiogenesis, aging, cancer, metabolic effects, etc.).

Existing assays (or others that can be developed on demand) can be applied to any cell-based on in vivo model in the lab

Indicative assays in cells

Tyrosinase activity (whitening), elastase activity, gelatin zymography for assessment of MMPs activity (anti-wrinkle), cell migration (wound repair), soft agar (anti-cancer activity), SA-β-gal and SenTraGor staining (cellular senescence), proteasome – lysosomal cathepsins enzymatic activity and in situ detection (anti-aging), ROS – proteome oxidation (oxidative stress – protection from oxidants, UV), cell viability assays (toxicity), imaging of lysosomes, mitochondria, aggresomes (proteome, mitochondria stability), mitochondrial respiration – functionality, gene cloning, siRNA – development of genes overexpressing cell lines, cell reporters for NRF2, NQO1 (anti-oxidant responses), NF-kB (inflammation), in vitro screening for 5-lipoxygenase (5-LO) inhibition, expression of major inflammatory modules (interleukins, TNF-α, NF-κB, COX-2 and 5-LO genes), cell-based estimation of nitric oxide (NO) production (inflammation), comet assay, detection of 8-oxo-dG, γΗ2Α.Χ (DNA damage), Q-RT-PCR, immunoblotting, angiogenesis assays (in human umbilical vein endothelial cells),expression of angiogenesis-related genes (e.g. VEGF), endothelial cells proliferation assay, tube formation, cell migration, matrix invasion, etc.

Indicative assays in flies

Study of proteostasis network: Autophagy-lysosome pathway (GFP-Atg8a, mcherry-Atg8a, Lamp-GFP, lysotracker, GABARAP staining) and in vivo proteasome – lysosomal cathepsins enzymatic activity. Study of heart structure (AtpA and phalloidin staining) and functionality (heart beat measurement). Detection of mitochondria structure/movement and of mitophagy process (ATP5a, MitoGFP, Mito-QC, Ref(2)p). Detection of oxidative status (roGFP transgenic lines for H2O2 or glutathione redox measurements, activation of oxidative response genes/Nrf2 targets (ARE-GFP lines), ROS levels, immunoblotting, detection of oxidized proteins. Metabolic analyses: mitochondrial respiration, lipids, glucose/trehalose levels. Aging and physiology assays: locomotor and memory analysis, resistance to stress, longevity, etc. Generation and in vivo pharmacological treatment of human disease models (neurodegeneration, cancer, heart dysfunction).

Study of molecular and cellular effects of drugs on peripheral blood mononuclear cells (PBMCs) and red blood cells (RBCs): Isolation of PBMCs and RBCs from whole blood (peripheral), proteasome activity, cathepsins activity (PBMCs), ROS levels measurement (PBMCs), confocal analysis (ATP5a – mitochondrial marker, aggresomes detection), immunoblotting, detection of oxidized proteins, gene expression (PBMCs), etc.
